PI3-K Assay Comparison Chart

AlphaScreen® technology is based on the emission of light (520-620nm) by Acceptor beads activated by the proximity of Donor beads. The interaction of biotinylated-PI(3,4,5)P3 and the PI(3,4,5)P3 binding protein brings both Acceptor and Donor beads together, producing a cascade of chemical reactions and leading to the amplified AlphaScreen® signal. This highly amplified signal is detected upon excitation of the Donor beads at 680 nm when singlet state oxygen (1O2) molecules are generated and diffuse to excite Acceptor beads. AlphaScreen® beads must be purchased seperately from PerkinElmer.

Class I Phosphoinositide 3-kinases (PI3-K), are ubiquitously expressed lipid kinases which phosphorylate PI(4,5)P2 at the 3'-hydroxyl of the inositol ring producing PtdIns(3,4,5)P3. PtdIns(3,4,5)P3 has shown to be an important lipid second messenger with key roles in fundamental cellular responses such as proliferation and survival. Due to these interesting biological roles, PI3-K and PtdIns(3,4,5)P3 have become attractive targets for research and drug development in many diseases including inflammation and cancer. Echelon has multiple formats available for the determination of class I PI3-K activity.
