Product Background: The Autotaxin Inhibitor Assay, developed by Echelon Biosciences Inc., uses the fluorogenic autotaxin substrate FS-3. FS-3 is an LPC analogue that is conjugated with both a fluorophore and a quencher. In its native state the quencher interferes with the fluorophore’s fluorescence. Once autotaxin cleaves FS-3, the fluorophore becomes liberated from the quencher, resulting in increased fluorescence.
Percent autotaxin inhibition is determined by referencing no inhibitor controls. The autotaxin inhibitor, BrP-LPA, is supplied to positively control for autotaxin inhibition. The assay is resistant to modest concentrations of DMSO and is performed in a continuous assay format, which has the advantage of being less susceptible to error arising from mis-timing the addition of components to different sample wells.
