Autotaxin (ATX) Activity Assay Service

The Autotaxin (ATX) Activity Assay Service uses the Autotaxin Activity Assay Kit (cat # K-4100) to quantify ATX activity in biological samples. ATX is an extracellular enzyme that cleaves lysophosphatidylcholine (LPC) to generate the phospholipid growth factor lysophophatidic acid (LPA). LPA is involved in a variety of biological functions, such as angiogenesis, wound healing, vascular and nervous system development, reproduction, pain transmission, immunological competence and the regulation of blood pressure. Altered LPA biology is associated with a number of diseases. Likewise, autotaxin has been implicated in a number of disorders, including a variety of cancers, obesity, arthritis, multiple sclerosis, Alzheimer’s disease and neuropathic pain.

Description

Assay Details

ATX activity is determined using the Autotaxin Activity Assay Kit (cat # K-4100). The assay utilizes the fluorogenic substrate, FS-3, an LPC analogue, that is conjugated with both a fluorophore and a quencher. When ATX cleaves FS-3, the fluorophore becomes liberated from the quencher, resulting in increased fluorescence. The assay gives a quantitative measurement of ATX activity in biological samples by measuring changes in fluorescence over time. 

Sample Considerations

The Autotaxin Activity Assay Service has been tested for the quantification of autotaxin activity in serum, plasma and cell culture media.

Sample Type: serum, plasma and cell culture media

Sample Volume: minimum of 500 μL of undiluted sample is needed

EDTA: Autotaxin is a metal dependent enzyme, avoid introducing EDTA or other metal sequestering compounds when collecting biological samples. For example, do not use EDTA tubes to collect plasma samples. EDTA will block ATX activity through metal ion depletion.

Cell culture samples: When using cell culture media it is important to grow your cells under serum-free conditions, since serum contains lyso-PLD activity. If this is not possible, please provide media that has not been in contact with cells as a control.

Storage and shipment of samples:

  • Freeze all samples before shipping and ship on 3-5 lbs of dry ice, if possible.
  • If samples cannot be frozen, use leakproof caps and vials.
  • Label vials well. If vials leak, the solution can remove poor labeling.
  • Please ensure all shipments containing human tissue or fluids are appropriately labeled as biohazard and packaged to prevent leakage during transport.
  • Contact your service coordinator and let them know the samples are on their way.

The minimum sample number for this service is 10. Example pricing is shown below and custom sample numbers can be accommodated.

Standard pricing is for each sample to be run in duplicate wells. This means that 20 samples = 40 assay wells. If you need to run greater or fewer samples, contact us for pricing.
