Boc-Val-Pro-Arg-AMC is a fluorogenic substrate for alpha-thrombin (Km = 21 μM, kcat = 109 s-1) and the thrombin-staphylocoagulase complex (Km = 25 μM, kcat = 89 s-1). The substrate is also cleaved by trypsin, tryptase from rat mast cells, Kex2 endoprotease, and acrosin and spermosin (trypsin-like enzymes from sperm of the ascidian Halocynthia roretzi).
Activity is quantified by release of free fluorescent 7-amino-4-methylcoumarin (AMC) which excites at 360-380 nm and emits at 440-460 nm.
